Studies on the metabolism of glycosaminoglycans under the influence of new herbal anti-inflammatory agents

The present study focused on the in vivo effect of an herbal-based, non-steroidal anti-inflammatory product boswellic acid on glycosaminoglycan metabolism. Glycosaminoglycan degradation was markedly reduced in all drug-treated animals as compared to controls.

Anti-arthritic activity of boswellic acids in bovine serum albumin (BSA) induced arthritis

The authors focused on the effect of boswellic acids on bovine serum albumin (BSA)-induced arthritis in rabbits. The local injection of boswellic acids (5, 10 and 20 mg) into the knee 15 min prior to the BSA challenge reduced the infiltration of leucocytes. Oral administration of boswellic acids (25, 50 and 100 mg/kg/day) significantly reduced the population of leucocytes in a BSA-injected knee. The results proved that the leucocyte-inhibitory activity of boswellic acids was not due to its cytotoxic effect.

Effect of a new non-steroidal anti-inflammatory agent on lysosomal stability in adjuvant induced arthritis

The present study explored the effect of non-steroidal anti-inflammatory agents on lysosomal stability. Adjuvant arthritic animals showed a significant increase in β-glucuronidase activity; a typical lysosomal enzyme, whose levels are elevated in arthritic syndrome. The increased rate of the release of β-glucuronidase from lysosome-rich fraction clearly suggested that arthritic syndrome caused decreased stability of the lysosomes. Administration of boswellic acids or salai-guggul to arthritic animals increased the lysosomal stability. Of the two anti-inflammatory agents tested, salai guggul was found to afford value that is more therapeutic.

Kinetic mechanism of guinea pig neutrophil 5-lipoxygenase

The current study investigated the kinetic mechanism of guinea pig neutrophil 5-lipoxygenase by spectrophotometric assay. Progress curves for reactions with arachidonic acid and eicosapentaenoic acid were characterized. Oxygenation of eicosapentaenoic acid in the presence of Ca2+ and ATP occurs with similar kinetics, except for significantly less substrate inhibition. This is the first report suggesting a kinetic mechanism for 5-lipoxygenase that accounts for substrate inhibition, regulated by Ca2+, ATP and substrate specificity.
